Sinister Rune relic in Castlevania: Belmont's Curse

Sinister Rune — Location and Effect

Quick answer: the Sinister Rune is a relic in the Clock Tower's Eastern Access Shaft. Fast-travel to the Mechanic's Ascent Mirror of Life, climb the gear shaft to the gated landing, then drop down the right wall beside the giant cog to a hidden ledge with a metal chest. Equipped, it adds 40% damage to a hit on an enemy that is still at full health — opening hits only. The 40% figure is guide-reported, not official. One guide prints 4.0%; every other source we checked says 40%, so 4.0% looks like a typo.

Is "rune" a separate system?

No. In Belmont's Curse the item category is Relics — passive equippables that sit alongside your weapon and Arcana. The Sinister Rune is the name of one relic, not a separate rune system. Across the guides we checked, no other item is called a rune.

Relics go into Rose's three relic slots (NoobFeed, All Things How, level C). The Sinister Rune does nothing while it sits unequipped in your inventory.

40% vs 4.0%

GameTrek prints "4.0% more damage". NoobFeed, All Things How, All Things How's relic tier list, Fanstanza and SportsRant all say 40%.

Four independent guides give 40% against one that gives 4.0%, and All Things How ranks the relic A-tier — a 4% opening-hit bonus would not support that ranking. Treat 40% as the working number and 4.0% as a likely typo. Neither figure has been measured in-game by us, and Konami publishes no relic stats.

Where to find the Sinister Rune

Both routes below describe the same chest in the Clock Tower's Eastern Access Shaft.

Route A — from the Mirror of Life (All Things How):

  1. Fast-travel to the Mechanic's Ascent Mirror of Life.
  2. Go right through Mechanic's Ascent and Tower Gallery into the Eastern Access Shaft.
  3. Climb the vertical gear shaft to the landing with the gated archway.
  4. Defeat the harpies, then move to the far right.
  5. Drop carefully along the right wall, past the rotating cog, onto the hidden lower ledge.
  6. Open the metal chest. Hold Y to equip immediately, or A to dismiss and equip later.

Route B — from inside the room (NoobFeed):

  1. In the vertical room, line up the two spinning platforms so both are flat and horizontal.
  2. Jump onto the upper platform.
  3. Slide beneath the wall opening into the machinery chamber.
  4. Clear the enemies and open the chest.

The two descriptions are not step-identical, but they agree on the room, the chest and the drop past the cog. The chest is off the main climb path — if you go straight up, you will miss it.

Is it worth a relic slot?

  • All Things How ranks it A-tier among relics (tier list, level C).
  • Fanstanza puts it fourth on its damage-relic ranking, behind Witch's Brew, Twilight Gem and Vile Trinket (Fanstanza, level C).
  • It rewards a single strong opening hit — bosses, elites, and the first enemy in a room. After that first hit connects, the relic contributes nothing until you meet a fresh full-health target.

Problems players hit

I climbed the shaft and never saw a chest. The chest is on a hidden lower ledge below the upper gated landing, beside the giant cog. Climb to the landing, go to the far right, and drop along the wall instead of continuing upward (All Things How, level C).

I picked it up but my damage didn't change. Equip it in one of the three relic slots. Inventory alone does nothing (All Things How, level C).

Does it work on bosses? Guides describe the condition as "an enemy at 100% health". We found no boss-specific test, so boss behaviour is unverified.

Does it stack with other damage relics? No source we read tests stacking. Unverified — do not assume the bonuses add together.

Is it 40% or 4.0%? See the comparison above. Four guides say 40%; one says 4.0%.
